The settlement was founded in 1660 by English colonists, named after Falmouth in Cornwall England, and incorporated in 1686. The town's 19th-century economy was based on whaling, agriculture (cranberries and strawberries), sheep husbandry, dairying, salt works, shipping.

The town encompasses several historic villages - Woods Hole, Falmouth Village Green, East Falmouth, North Falmouth, West Falmouth, Teaticket, Hatchville, Waquoit. In Woods Hole are located several scientific organizations and marine facilities (Oceanographic Institution, Marine Biological Laboratory, Woodwell Climate Research Center, Science Aquarium, museums).
